Conjugation of trastuzumab variants and PE24-AzF. Analysis of SDS-PAGE was done under a non-reducing condition for reduced, reoxidized, and PE24-conjugated trastuzumab. The reduced/reoxidized trastuzumab were first conjugated to DBCO-polyethylene glycol (PEG4)-maleimide using the thiol-maleimide coupling reaction. The modified antibodies were then reacted with PE24-AzF via the strain-promoted azide-alkyne cycloaddition reaction
